Trying To Extract Deadly Plant Alkaloids

In todays video I will try to extract the deadly tropane alkaloids from datura stramonium. This plant contains atropine, scopolamine and hyoscyamine. As all of these alkaloids are deadly you should not repeat this. We might have failed to obtain pure crystals of said alkaloids but the finished extract is extremely toxic anyways.

I don't think you would have destroyed the alkaloids, the fluffy flocculant in the ether looked great. Try allowing the particles to aggregated a little and maybe cool the ether solution down to kick out as much of the compounds as possible and then filter through a coffee filter when the particle size has increased a little. You definitely need to do either a filtration or a centrifugation step here to separate all the other junk that has stayed dissolved in the ether, you wont be able to just boil it off.

Please be careful if you do decide to centrifuge stuff in ether. It will explode if it sparks and a spark-proof centrifuge should be used.

The HCl salt in ether just needed filtering. Heating it probably was a mistake, tropanes can get tricky like that. Cooling the mix in the freezer and filtering would been ideal, then you just dry it at room temp. The fluffy suspension is sexy af by the way.

Cool video. I extracted nicotine, mo hung in different solvents. I have used isopropyl alc and pyrrolodine to extract stychnine and brucine. For safety I only extracted 1.8 g of nux vomica. That 20 to 35 mg of Strychnine and 28 mg brucine. Sodium dichromate and hcl turned the solution is dark purple.

First step grind up and dry absolutely in z sealed container that passes air over dry mgso4. 2nd step dry your hexzne, etc, and make it acidic by bubbling HCL gas through it. Then, load into your soxlet extractor. Add this into your reveiver and just enough to soak the seed powder. The alkaloids will absorb some hcl the rest will off gas. Let it run 4 to 6 hours at aggressive reflux. This removes oils and fats. The hexane wash can be discarded. Next, add a bit of Ca(OH)2 on top of the seed mash, and repeat the procedure with ammonia saturated DCM to get alkoids as a free base. Distill off the DCM Clean up can be done by a simple acid base extraction, aim for PH of 2 to 3 for the acid step and 12 to 13 for the base step. Diethyl ether is great for the organic pulling in this step. Weigh the flask so you can calculate the amount of alkaloids present. Add the eter solution of alkaloids and distill off ether weigh again to get your alklod amount and dissolve in anhydrous isopropyl alcohol and make a solution of sulfuric acid in isopropyl alcohol in slightly more than equimolar amount and mix. Your alkaloids will crash out.❤

Yeah like 98% of them trip reports end up in a hospital visit ... My friend once took it too, he was kinda lucky as never badly physically hurt himself, but I'm pretty sure suffered some major mental trauma from it for several years following his "experiment". Truly scary stuff.

I've done brugmansia in a controlled setting. It was a hauntingly beautiful experience. Losing the ability to speak or put objects down reveals you for who you truly are deep down on the inside. Waking up to people who love me around me taught me that I had nothing to be afraid of in this world and that without recognition and validation I would be an empty observer. I have experience with alkaloid isolation. Seeds contains huge amount of fat so you should did extraction with pure solvent (hexane or heptane without amonia) first to get rid of fat. It easier to use leafs (without fats). Tropane alkaloids are easy extractable and there is ni need for Soxhlet (these alkaloids are esters so there is some decomposition with hot amonia). Extraction with warm acid water is enough. After that add ammonia and extract into organic solvent.

Yup. Years ago I tried extracting what I thought it penicillin from some moldy coconuts using DCM, but instead of getting a product I ended up with a weird ugly butter, and had no way of removing the fat, so had to destroy my product, since it was useless.

The extractor is a continuous liquid/liquid extractor. It is intended to extract things from light liquids (hexanes are common) into water but can also be used to extract things from water into chloroform or other heavy liquid.

It can extract things into a liquid with a poor or even inverse partition coefficient. Ie. Equally or more soluble in the lighter liquid.

Tips for less impurities: acidify the product and wash with nonpolar. then freebase product, wash with water.
When removing solvent, avoid heat. maybe try reduced pressure.

Use diethyl or even subcritical cold methyl ether as a solvent and then tartaric acid to salt when dealing with delicate alkaloids. HCl, H2SO4 and Heptane and Dichloronethabe are too harsh. You could try guanidine as the alkali to freebase, it's milder than Ammonia thats why its used in mild afro hair relaxers.

Chances that an akaloid dissolves in hexane are lousy. I would powderize the seeds and then do test extractions with THF or Ethylacetate or CHCl2 on just a gram of the seeds. Then run a TLC to check if the extraction works. In the first place read up on the available literature!

Scopolamine has been known to be used to 'knock out' unsuspecting persons so they can be robbed. I believe I first read of this being used in Colombia.

@@z2kk in a soxhlet extractor, the tube on the side is used as a siphon: the main chamber with the things to extract fills up gradually because of the condensation of the vapours of the solvant, and when it's filled enough the whole chamber is emptied of its solvant: that's one cycle.

Here, the tube that could allow the cycling is pressure equalised to the rest of the apparatus, meaning that it can't cycle: it works continuously
